What Eloralintide is
Eloralintide (LY3841136) is Eli Lilly's investigational selective amylin receptor agonist, engineered to activate the AMY1 receptor with approximately 12-fold greater potency than the calcitonin receptor. Amylin is a pancreatic satiety hormone co-secreted with insulin; by mimicking its action in appetite-control pathways of the hypothalamus, eloralintide increases feelings of fullness, reduces caloric intake, and slows gastric emptying — the three central pillars of amylin-driven weight regulation.
Unlike earlier amylin analogs, eloralintide's receptor selectivity is intended to preserve efficacy while improving tolerability, particularly reducing the nausea and vomiting commonly seen with non-selective agonists.
Because it is a selective amylin agonist, eloralintide is mechanistically distinct from GLP-1 agonists (semaglutide, tirzepatide) and can be modelled as a complementary — not redundant — component of a metabolic protocol. It should NOT be combined with cagrilintide, which activates the same receptor and would produce competitive binding rather than synergy.
